《死亡之屋》作为一款光枪射击游戏，玩家以使用光枪（在PC中则为鼠标）瞄准和射击画面上的的丧尸来击退它们。主角的手枪使用可容纳6发子弹的弹匣；玩家通过射击屏幕外部来重新填弹。当玩家受到伤害或射击人质时，会扣除生命值，失去所有生命值时会出现询问继续画面。如果玩家耗尽了继续次数，则游戏结束。整个游戏过程中提供急救包，可以恢复生命值；有些可以从获救的人质中获得，而另一些则隐藏在场景中的可击碎物品中。特殊物品亦可在可击碎物品中找到，向击破它们的玩家提供奖励。玩家可以在每个关卡结算时，根据获救的人质数量，获得额外的恢复生命值。
在整个游戏过程中，玩家会面临许多情况，玩家的决策将对游戏的前进方向产生影响。如在游戏首关，人质即将从桥上被怪物扔下而死。如果玩家救了人质，路线将直接从前门进入房屋；但是如果玩家未能营救人质，路线将被设定为向下穿越下水道。如果玩家解救了所有人质，一个充满生命和奖金的秘密房间会在游戏结束时出现。玩家亦可以通过射击敌人头部和解救人质来获得额外积分。
